Sigma Life Science launches suite of knockout rats to model Parkinson’s disease

Sigma Life Science, the biological products and services research business of Sigma-Aldrich, announced that its SAGE Labs initiative has created a suite of knockout rat models of Parkinson’s disease in collaboration with The Michael J Fox Foundation for Parkinson’s Research (MJFF). Developed using the proprietary SAGEspeed model creation process, these off-the-shelf rat models represent a breakthrough in Parkinson’s disease research, overcoming the inherent limitations of the existing mouse models.

Knockout rat models offer significant advantages over the existing mouse models, which do not effectively mimic the symptoms of Parkinson’s disease in humans. The neuronal circuitry of the rat model more closely imitates that of humans, better demonstrating the disease symptoms, and rats’ size and behaviour allow researchers to perform a variety of experimental procedures more easily than in mice. In collaboration with MJFF, SAGE Labs has used Sigma Life Science’s CompoZr Zinc Finger Nuclease (ZFN) technology to create a range of knockout rat models specific to Parkinson’s disease, based on existing knowledge of the key genes implicated in the disease. This groundbreaking collection of rat models currently includes LRRK2, Parkin, Pink1 and DJ-1 knockouts which will help researchers accelerate investigation of novel therapeutic strategies for Parkinson’s disease.

Sigma adds 1,400 cell lines to its US portfolio of ECACC products

Sigma Life Science, the biological products and services business of Sigma-Aldrich, has announced the addition of 1,400 new European Collection of Cell Culture (ECACC) cell lines to its collection of cell line cultures and services. These cell lines have been fully authenticated and tested free of mycoplasma and can now be delivered within days to customers in the U.S., Mexico and Canada. ECACC is a recognized worldwide expert in the maintenance, cultivation and distribution of authenticated cell lines. The collection of cell lines can be accessed at www.sigma.com/ecacc.

Scientists rely upon these cell lines, which include human, mouse and insect, for cell-based assays that determine the safety and efficacy of drug candidates. Due to high costs and the great amount of time required for research and development, drug developers prefer to use cell-based assays in cell signaling studies, cancer research and other areas in order to correct issues that may arise, as early as possible, in the research cycle.

Oncology disease model cell lines

Sigma Life Science, the biological products and services research business of Sigma-Aldrich, today announced the addition of new genetically modified cancer cells to the company’s range of high-quality cell lines. These human knockout and knock-in oncology cell lines, created using Sigma’s proprietary CompoZr Zinc Finger Nuclease (ZFN) technology which received a silver medal as a Top Ten Innovations of 2010 from The Scientist magazine, offer researchers cellular models of cancer that are expected to enhance development of drugs for personalized medicine.

Free public database of exclusive genomic information

Sigma Life Science, the biological products and services business of Sigma-Aldrich, today announced that its SAGE Labs initiative has partnered with Cofactor Genomics in an effort to sequence genomes for six of the most widely used strains of rat. As part of the agreement, Cofactor Genomics plans to generate and analyze sequence data using next generation sequencing systems, while SAGE Labs plans to provide the samples and to host a new, free public database at sageresearchmodels.com in late 2011. The database will include exclusive genomic information and analysis tools.

Knock-in rats using zinc finger nuclease technology

Sigma Life Science, the biological products and services business of Sigma-Aldrich, today announced that its SAGE Labs initiative has successfully used its proprietary CompoZr Zinc Finger Nuclease (ZFN) technology to generate knock-in rats in which foreign genes have been inserted, or ‘knocked-in’, into the rat genome in a precisely targeted manner. This breakthrough achievement represents a major step forward in the creation of a transgenic animal, which may serve as more predictive models of human disease.

In a proof of concept study conducted by researchers at SAGE Labs, a copy of the green fluorescent protein (GFP) was inserted in a targeted fashion into the Mdr1a gene of a rat, thus creating a rat that glows under special lighting conditions. The study, which has recently been accepted for publication1, opens the doors to a variety of new applications including the tagging of genes with reporters such as GFP to measure gene expression patterns and replacement of rat genes with humanized versions.

Joint project to develop microRNA target validation system

Sigma Life Science, the  biological products and services research business of Sigma-Aldrich, and SwitchGear Genomics, Inc have announced a joint project to develop and distribute a novel microRNA (miRNA) target reporter system.

Combining SwitchGear Genomics’ expertise with Sigma’s lentiviral technology, these ready-to-use reporter vectors are expected to simplify miRNA target validation, enabling researchers to investigate gene regulation in a variety of biological pathways.
